摘要： Alzheimer's disease constitutes a progressive brain-destroying disorder that leads people to lose memory abilities while experiencing behavioral modifications and gradual mental deterioration. The worldwide population affected by AD stands at 50 million individuals. The current treatments provide symptomatic relief instead of therapeutically treating the altered neurological processes. Plant-derived compounds have demonstrated promising neuroprotective effects by modulating key pathways implicated in neurodegenerative diseases. The present research evaluated the potential of desmethylglycitein against scopolamine-induced cognitive impairment in mice. Desmethylglycitein is a naturally occurring isoflavone, primarily found in soy products and other leguminous plants. The research period lasted for 9 days, during which desmethylglycitein (10, 20 and 40 mg/kg) or donepezil (5 mg/kg) was administered. Scopolamine (2 mg/kg) was given on days 7-9. Behavioral tests (MWM, Y-maze and OFT) were performed on days 7 and 9, followed by biochemical analyses. Scopolamine treatment significantly impaired memory and locomotor activity. The memory impairment caused by scopolamine was significantly alleviated by desmethylglycitein, as evidenced by reduced escape latencies in MWM and increased percentage of spontaneous alternation in Y-maze. Desmethylglycitein treatment also improved locomotor activity in the OFT as evidenced by an increased number of square crossings. In the biochemical analysis, scopolamine treatment significantly decreased brain GSH levels and increased brain TBARS levels. Furthermore, it reduced serum Nrf-2 levels and elevated serum IL-1 beta levels. Pretreatment with desmethylglycitein significantly mitigated these biochemical changes induced by scopolamine. This suggests that desmethylglycitein may have the potential to counteract scopolamine-induced deficits in memory and locomotion, possibly by inhibiting oxidative and inflammatory stress.

摘要： BackgroundDigestive perianastomotic ulcerations (DPAU) are rare Crohn's disease (CD)-like lesions that develop following intestinal surgery. Although enteral nutrition and anti-inflammatory agents constitute first-line therapies, 59% of refractory cases require advanced interventions. Emerging evidence implicates tumor necrosis factor (TNF)-alpha-driven inflammation as a key pathogenic mechanism, yet optimal therapeutic strategies remain *** summaryA 12-year-old girl with a surgery history of neonatal necrotizing enterocolitis (NEC) presented with severe anemia (hemoglobin: 51 g/L) and chronic abdominal pain. Endoscopy revealed circumferential ulcers at the ileocolonic anastomosis, histologically confirmed as active inflammation without granulomas or evidence of infections. Initial therapy with 3 months enteral nutrition and mesalamine failed. Subcutaneous adalimumab induced endoscopic ulcer healing and hemoglobin normalization (111 g/L) within 3 months. At the 12-month follow-up, sustained clinical remission was observed (hemoglobin: 120 g/L, fecal calprotectin: 45 mu g/g), with complete mucosal healing and no adverse *** anti-TNF therapy induced rapid and sustained remission in a pediatric patient with DPAU, reinforcing the mechanism that TNF-alpha-driven inflammation underlies disease pathogenesis. While promising, these findings require validation in larger cohorts. Individualized therapeutic decisions should weigh the risks of recurrence, cost, and long-term biologic safety.

摘要： 年龄相关性黄斑变性(AMD)是一种慢性视网膜退行性疾病,严重损害患者的中心视力。研究表明,补体系统与AMD的发生和发展密切相关,在其发病机制中扮演着关键角色,涉及遗传风险、炎症反应和氧化应激等多种因素。本文就补体系统的激活与AMD的关系及近年来靶向补体治疗AMD的临床试验进行综述,以期为AMD的基础研究和临床治疗提供参考。

摘要： 目的:探讨三氧化矿物凝聚体(Mineral Trioxide Aggregate,MTA)对年轻恒牙部分活髓切断术患儿血清白介素-1β(Interleukin-1β,IL-1β)、碱性成纤维细胞生长因子(Basic fibroblast growth factor,bFGF)水平及治疗成功率的影响。方法:选取2022年3月-2023年9月期间在我院口腔科实施年轻恒牙部分活髓切断术患儿109例(共109颗牙)作为研究对象。随机将研究对象分为对照组(n=54)和观察组(n=55)。两组患儿均进行年轻恒牙部分活髓切断术。在此基础上,两组采用不同的术中盖髓剂。对照组采用氢氧化钙作为术中盖髓剂。观察组采用MTA作为术中盖髓剂。分析评估两组的治疗成功率,牙周指数[牙周袋深度(Periodontal Pocket Depth,PD)、评估牙龈指数(Gingival Index,GI)、龈沟出血指数(Sulcus Bleeding Index,SBI)以及菌斑指数(Plaque Index,PLI)],血清IL-1β、bFGF水平以及不良反应发生情况。结果:两组术后3 m的治疗成功率比较无差异(P>0.05),观察组术后12 m的治疗成功率显著高于对照组(P<0.05)。术后3 m,两组PD、GI、SBI、PLI均比术前显著降低,且观察组的PD、GI、SBI、PLI均显著低于对照组(P<0.05)。术后7 d,两组IL-1β水平均比术前显著降低、bFGF水平均比术前显著提高,且观察组的IL-1β、bFGF水平的变化幅度均显著大于对照组(P<0.05)。结论:与氢氧化钙相比,MTA可提高年轻恒牙部分活髓切断术患儿治疗成功率,促进牙根发育,调节血清IL-1β、bFGF水平,且具有一定的安全性。

摘要： 目的:探讨IL-17在大鼠高血压心力衰竭中的作用及机制。方法:自发性高血压(SHR)大鼠采用腹主动脉套扎方式建立高血压心力衰竭模型,采用外源性注射将IL-17+IgG和IL-17蛋白注射至高血压心力衰竭大鼠腹腔内,4周后,监测心脏结构与功能,并收集外周血及心肌组织,采用HE染色、免疫组化、Western blot、qRT-PCR、ELISA方法研究IL-17在高血压心力衰竭中的作用及机制。结果:外源性腹腔注射IL-17后,提高了IL-17、NT-proBNP水平、P38和MK2蛋白表达,降低了VEGF水平和Cav1.2蛋白表达。外源性腹腔注射IL-17+IgG阻断了IL-17,提高了VEGF水平和Cav1.2蛋白表达,降低了NTproBNP水平、P38和MK2蛋白表达。结论:IL-17可激活P38/MK2-Cav1.2信号通路参与大鼠高血压心力衰竭及心功能损伤;抑制IL-17可有效改善高血压心力衰竭造成的心功能损伤。

摘要： 目的探讨白细胞介素-6(IL-6)在脓毒症并发急性肺损伤(ALI)患者中的表达及其与预后的相关性。方法选取北京市大兴区中西医结合医院于2021年2月—2024年2月收治的111例脓毒症并发ALI患者为研究对象。根据28 d生存状况将患者分为生存组(73例)和死亡组(38例),另选健康体检者68名作为对照。记录临床资料,检测受试者的血清C-反应蛋白(CRP)、肿瘤坏死因子-α(TNF-α)等实验室指标,进行脓毒症并发ALI患者死亡的危险因素分析,并验证各指标对脓毒症并发ALI患者预后价值。结果对照组、低危组、中危组、高危组IL-6分别是(36.48±13.50)、(114.17±11.67)、(168.52±15.80)、(209.43±20.77)pg/mL,TNF-α水平分别是(47.29±12.29)、(66.55±9.94)、(72.96±10.47)、(80.43±10.88)mg/L,低危组、中危组、高危组IL-6、TNF-α水平高于对照组,且病情越重,IL-6、TNF-α水平越高(F=1190.96、85.021,均P<0.05)。死亡组脓毒症分级为高危比例42.11%(16/38),生存组脓毒症分级为高危比例21.62%(16/74);死亡组机械通气比例92.11%(35/38),生存组机械通气比例70.27%(52/74);死亡组脓毒症分级为高危、机械通气比例高于生存组(脓毒症分级χ^(2)=8.517,机械通气χ^(2)=6.904,均P<0.05)。死亡组和生存组CRP水平分别是(46.62±4.77)、(43.79±4.82)mg/L;血乳酸平分别是(3.89±1.52)、(3.21±1.07)mmol/L;IL-6水平分别是(202.79±38.19)、(128.58±35.64)pg/mL;TNF-α水平分别是(78.74±10.23)、(69.59±12.81)mg/L,死亡组CRP、血乳酸、IL-6和TNF-α水平高于生存组(t=2.952、2.748、9.955、3.819,均P<0.05)。脓毒症分级为高危、进行机械通气、CRP、血乳酸、IL-6和TNF-α高水平均是脓毒症并发肺损伤患者死亡的危险因素(P<0.05)。ROC曲线结果表明,IL-6预测预后的曲线下面积达到0.919,灵敏度为89.5%,特异度83.8%。结论血清IL-6水平在脓毒症并发ALI患者中显著升高,并且与患者的预后密切相关。IL-6可以作为评估脓毒症并发ALI患者预后的潜在生物标志物,为临床治疗提供参考。
