摘要： Globally, colorectal cancer (CRC) is the primary cause of cancer-related fatalities. Our previous study demonstrated the efficacy of Jianpi Lishi Jiedu Granules (JLJG) in preventing postoperative recurrence of colorectal adenoma (CRA). Building on this foundation, the current study aims to elucidate whether the mechanism by which JLJG prevents postoperative recurrence of CRA involves the classical JAK/STAT inflammatory signaling pathway and to assess its specific impact on this pathway. Utilizing proteomics, we discerned 143 differentially expressed proteins (DEPs) regulated by JLJG, whose functional roles are intimately linked to the JAK/STAT signaling pathway. Among these, we identified key proteins such as IL-6, JAK1, JAK2, STAT3, CCND1, MYC, BclXL, and SOCS3 that are regulated by JLJG and play pivotal roles in the JAK/STAT signaling cascade. Our findings indicate that the sustained activation of the IL-6/JAK/STAT3 signaling pathway is significantly associated with CRA recurrence. JLJG was found to effectively modulate the expression levels of these proteins, as well as the expression of downstream genes including BCL2, MCL1, P21, and JAK1, STAT3, thereby inhibiting the IL-6/JAK/ STAT3 signaling pathway. Consequently, this study demonstrates that JLJG prevents the postoperative recurrence of CRA by inhibiting the IL-6/JAK/STAT3 signaling pathway and its negative feedback loops.

摘要： Background. The development of de novo donor-specific antibodies (dnDSA) and antibody-mediated rejection (AMR) remains a barrier to long-term graft and patient survival. Most dnDSA are directed against mismatched donor HLA-DQ antigens. Here, we describe a novel algorithm, which we have termed categorical amino acid mismatched epitope, to evaluate HLA-DQ mismatches. Methods. In this algorithm, amino acid residues of HLA-DQ protein were categorized into 4 groups based on their chemical characteristics. The likelihood of categorically mismatched peptides presented by the recipient's HLA-DRB1 was expressed as a normalized value, %Rank score. Categorical HLA-DQ mismatches were analyzed in 386 heart transplant recipients who were mismatched with their donors at the HLA-DQB1 locus. Results. We found that the presence of DQB1 mismatches with %Rank score <= 1 was associated with the development of dnDSA (P = 0.002). Furthermore, dnDSA increased the risk of AMR only in recipients who had DQ mismatches with %Rank score <= 1 (hazard ratio = 5.8), but the freedom from AMR was comparable between recipients with dnDSA and those without dnDSA if %Rank scores of DQ mismatching were >1. Conclusions. These results suggest that HLA-DQ mismatches evaluated by the categorical amino acid mismatched epitope algorithm can stratify the risk of development of dnDSA and AMR in heart transplant recipients.

摘要： PURPOSE. To investigate the ocular surface changes during progress of the Sj & ouml;gren's Syndrome (SS), using a previously described IL14 alpha transgenic mice (IL14 alpha TG) SS model. METHODS. The ocular surface of IL14 alpha TG and C57BL/6 wild-type (WT) female mice were evaluated at the age of six, nine, 12, 15, and 18 months. Slit lamp microscopy observation, Oregon green dextran staining, Schirmer test, and periodic-acid-Schiff staining were assessed. Immunohistochemistry, immunofluorescence, and associated gene expression analysis by qPCR and ELISA were performed in cornea, conjunctiva, and lacrimal grand at different ages of the mice. Masson's trichome staining was conducted on lacrimal gland cryosections. RESULTS. Compared with C57BL/6 WT mice, IL14 alpha TG mice showed corneal barrier function damage and losses in conjunctival goblet cell density starting at nine months, whereas decreases in tear secretion started at 18 months of age. Significant increases in CD4+ T cell infiltration in the conjunctiva of IL14 alpha TG mice was first observed at 6 months. Higher expression levels of inflammatory cytokines IL-17A, IFN-gamma , IL-1 beta, and TNF-alpha in the conjunctiva, whereas MUC5AC and MUC5B had lower expression levels at nine months in the IL14 alpha TG mice. However, lacrimal gland function-associated gene expression levels mostly decreased in IL14 alpha TG mice at 12 months of age. CONCLUSIONS. Ocular surface tissue changes were involved in SS-like dry eye in a time-dependent manner in IL14 alpha TG mice, and conjunctival T-cell infiltration may contribute to ocular surface pathological changes in an early stage of SS-related dry eye.

摘要： Background: In human tuberculosis, Mycobacterium tuberculosis elicits immunopathology characterized by abnormal expression of plasma cytokines. This immunopathology may influence the severity of illness and the efficacy of prolonged anti-mycobacterial treatment. Methods: The study analysed TNF-alpha, IL-10, and TGF-(3 levels in 90 pulmonary tuberculosis patients and compared them with 90 healthy individuals. The tuberculosis patients were categorized as mild, moderate, or advanced based on chest x-ray results, and the ratio of pro-inflammatory to anti-inflammatory cytokines was compared. Results: All three cytokines were present in most tuberculosis patients, and their concentrations were noticeably higher than in healthy individuals. The ratio of TNF-alpha/IL-0 and TNF-alpha/TGF-(3 demonstrated the most powerful discriminatory ability in determining the severity of tuberculosis. The levels of TNF-alpha/IL-0 and TNF-alpha/ TGF-(3 in individuals with moderate and advanced tuberculosis were markedly elevated compared to those with mild cases. The area under the curve (AUC) for TNF-alpha/TGF-(3 and TNF-alpha/IL-0 was determined to be 0.823 and 0.814, respectively. Conclusions: The elevated ratio of TNF-alpha/IL-0 and TNF-alpha/TGF-(3 in the sera of TB patients may contribute to an excessive pro-inflammatory response, whereas a lower ratio may regulate excessive inflammation. Both the TNF-alpha/IL-0 and TNF-alpha/TGF-(3 ratios may effectively categorize the tuberculosis severity and have the potential to serve as biomarkers for evaluating the early response to treatment.

摘要： Background: : Older adults experience chronic inflammation, which is associated with health conditions such as sarcopenia, and resulting in reduced functional capacity. Resistance training (RT) is a beneficial intervention for improving health in the elderly. Objective: : This study aimed to investigate the effect of RT on inflammatory biomarkers, body composition, and functional capacity in healthy adults aged 60 years and over. Additionally, this study conducted a metaregression to investigate the moderating effect of exercise variables on inflammatory markers. Method: : Medline, PubMed, Web of Science, Cochrane Library, Embase, and Google Scholar were searched systematically until December 2023. Randomized controlled trials (RCTs) assessing the impact of RT on Creactive protein (CRP), interleukin-6 (IL-6), and tumor necrosis factor-alpha (TNF-alpha), body weight, BMI, fat percentage, fat mass, lean mass, leg extension strength and six-minute walk test (6-MWT) were included. Effect size was estimated by using the mean difference (MD) with 95 % confidence interval (CI). Results: : Nineteen RTCs involving 728 participants were included. The results revealed that CRP levels significantly decreased following RT programs (MD:0.74, p = 0.008), while TNF-alpha (MD: 0.1, p = 0.95) and IL-6 (MD:0.27, p = 0.12) did not show significant changes. Additionally, RT enhanced leg extension strength and 6-MWT performance. Conclusion: : RT effectively reduces CRP concentrations and enhances functional capacity in healthy older adults. However, it does not have a significant impact on TNF-alpha and IL-6 levels. Future researches are needed to make a clear conclusion and understand the mechanisms underlying the effects of RT in healthy older adults.
